Title: Zymogram Polymorphism of Camphor Silkworm Larval Esterase Isozyme and Its Application in Analysis of Genetic Relationship Between Varieties
Abstract: Camphor silkworm(Dictyoploca japonica) is a rare silkworm resource.In present study,the zymograms of esterase isozymes during different developmental stages and in various tissues and organs of camphor silkworm larvae were determined using polyacrylamide gel electrophoresis.Moreover,the genetic relationship between two varieties named Chunhei and Chunlu was analyzed based on polymorphism of the esterase isozyme zymograms.The results showed that the activity of esterase isozymes and the number of enzymatic bands from midgut of camphor silkworm larvae increased with the advance of larval development,and the highest expression of isozymes was at the full appetite stage within the same instar.Higher activity of esterase isozymes and higher number of enzymatic bands were observed in midgut and fat body of the larvae.Zymograms of esterase isozymes from the two tested varieties were obviously different.Analysis on difference of the zymograms yielded a genetic similarity coefficient of 0.86 between the two varieties,showing relatively close genetic relationship.The obtained results suggest that the activity of esterase isozymes and number of enzymatic bands are related to growth,development and tissue functions of the camphor silkworm larvae.Therefore,esterase isozyme zymogram of camphor silkworm larvae can be used as a reference to analyze genetic relationship between different varieties.
