Title: Content Determination of Ursolic Acid and Oleanolic Acid in Comus Officinalis by High Performance Liquid Chromatography
Abstract: Objective To determine the content of ursolic acid and oleanolic acid in Comus officinalis by high performance liquid chromatography(HPLC) and to provide a basis for the quality control of Comus officinalis.Methods HPLC was performed on a Diamonsil C18 column(4.6 mm × 250 mm,5 μm) at a column temperature of 25 ℃,a flow rate of 1.0 ml/min,a sample size of 20 μl,and a detection wavelength of 210 nm.The mobile phase used in HPLC consisted of water and a mixture(composed of water,glacial acetic acid,and triethylamine at a ratio of 30∶0.1∶0.05) at a ratio of 88∶12.Results Ursolic acid and oleanolic acid had a good linear relationship between concentration and peak area in the concentration ranges of 0.031 5-0.315 mg/ml and 0.015 6-0.156 mg/ml,respectively;the average recovery rates of ursolic acid and oleanolic acid were 97.53%(RSD = 1.28%) and 100.08%(RSD = 1.98%),respectively.Conclusion This HPLC method is easy to operate,highly repeatable,accurate,and reliable and can be used for the quality control of Comus officinalis.
