Title: Inhibited proliferation of WE-68 cell line induced by flavopiridol in vitro and in vivo
Abstract: [Objective] To study the mechanism of growth inhibition induced by flavopiridol in human Ewing Sarcoma cell line WE-68. [Methods] Inhibitory effect on growth was observed by MTT method. Apoptosis of WE-68 cells was dectected by DNA ladder and flow cytometry. Gene (bcl-2, bcl-xl, bax, mcl-1) expressions related with apoptosis were examined by immunoblotting method. The subcutaneous tumors of SK-N-MC cells were established in nude mice to study the antitumor effects of FP; TUNEL staining was examined to study weather FP induces apoptosis in vivo. [Results] Flavopiridol could inhibit the growth of WE-68 cells significantly, related with concentration and time. Apoptosis was abserved in WE-68 cells treated with flavopiridol in vitro and in vivo. Flavopiridol could decrease mcl-1 protein expression, but could not affect bcl-2, bcl-xl and X-IAPprotein expression. [Conclusions] Flavopiridol could inhibit the growth of WE-68 cells and induce apoptosis in vitro and in vivo. We proposed that its mechanisms were probably associated with decreased mcl-1 protein expression.
