Title: Immobilized glucoamylase with sodium alginate and gum arabic and its properties
Abstract: Using sodium alginate(SA) and gum arabic(GA) as the carrier to prepare immobilized glucoamylase, on the basis of single factor experiment, using enzyme activity recovery rate as evaluation indexes, the optimal conditions for glucoamylase immobilization were optimized by response surface methodology. The optimal conditions were SA-GA mass ratio 2.7 ∶1, calcium chloride concentration 6.2 g/100 ml, and immobilization time0.8 h. Under these conditions, the immobilized enzyme activity recovery rate was up to 67.91%. The enzymatic properties of the immobilized enzyme were studied, and the results showed that the optimum reaction p H and temperature of immobilized enzyme was the same as the free enzyme of p H4.6, temperature 45 ℃, and both the thermal stability and operational stability of the immobilized enzyme were better than the free enzyme.
