Title: Studies on Immobilizing α-amylase with Sodium Alginate
Abstract: Using sodium alginate as carrier, α-amylase was immobilized by the embedding-crosslinking method. The optimal conditions were determined with the single factor experiment and the orthogonal experiment. The immobilized enzyme with porous structure was prepared by using polyethylene glycol 4 000 as the porogenic agent. The results showed that the optimal conditions were 1.6% sodium alginate, 0.4% α-amylase and 0.2% sodium carboxymethyl cellulose, CaCl2 2%. Under the optimal conditions, the immobilization efficiency was 92.43%. The enzyme activity could be improved by 37.61 percentage with 2.5% PEG 4 000 in the optimized system. The temperature and the pH stability of the immobilized enzyme were found to be better than that of the free enzyme.
