Title: Determination of 4 flavonoids and fingerprint study in Rehe Scutellaria baicalensi by HPLC
Abstract: OBJECTIVE To develop an RP-HPLC method for the quantitative analysis of baicalin,scutellarin,and establish a fingerprint of flavonoids in Rehe Scutellaria baicalensi.METHODS The methanol extract of genuine Scutellaria baicalensi was separated on a Diamonsil C18(4.6 × 150 mm,5 μm) gradient elution using acetonitrile and H2O-acetic acid-triethylamine(100:0.4:0.1) solvent system and detected at 280 nm.Flow rate was 1.0 mL·min-1 and 10 μL was injected every time.RESULTS The linear range of scutellarin,baicalin,baicalein and wogonin were 2.50-80.00 μg·mL-1(r = 0.9999),10.78-345.0 μg·mL-1(r =0.9999),3.50-112.0 μg·mL-1(r =0.9998),2.00-64.00 μg·mL-1(r =0.9998),respectively.The average recovery rates of scutellarin,baicalin,baicalein and wogonin were 99.1%(RSD = 1.7%),100.4%(RSD = 1.5%),99.2%(RSD = 1.8%),99.8%(RSD = 2.2%),respectively.In the fingerprint,12 common peaks of flavonoids in Rehe Scutellaria baicalensi were confirmed.CONLUSION The methods can be used scientifically to evaluate the quality of Rehe Scutellaria baicalensi both qualitatively and quantitatively.
