Title: Gastrodin Represses Expression of Cytokine Induced by Homocysteine in Microglia
Abstract: Objective: To evaluate the effect of gastrodin on the expression of interleukin-1β( IL-1β),interleukin-6( IL-6) and tumor necrosis factor-α( TNF-α) in culturing for microgia( BV-2 cells) induced by homocysteine and then to compare the effect with that of EGb761. Method: The BV-2 cells incubated vitro were divided into: blank control group,homocysteine( Hcy) group,EGb761 group( 100 mg / L) and gastrodin groups of different concentration( 2. 5,5,10 mmol/L). The mRNA expressions of IL-1β,IL-6 and TNF-α were assessed by RT-qPCR. The supernatant protein was detected by ELISA. Result: The mRNA expression and supernatant protein of IL-1β,IL-6 and TNF-αin Hcy group were significantly higher than those of control group,but were decreased significantly in EGb761 group and gastrodin groups. Especially,both the mRNA expression and supernatant protein in higher concentration gastrodin group were comparable to those in EGb761 group. Conclusion: Gastrodin can inhibit the expressions of IL-1β,IL-6 and TNF- α in cultured BV- 2 cells induced by homocysteine. In a certain concentration,it was less effective than EGb761.
