Title: Effect of Amyloid-β Protein on Intracellular Calcium Concentration in Cultured Hippocampal Neurons
Abstract: Objective To observe the effect of amyloid β protein(Αβ) on intracellular calcium concentration in cultured hippocampal neurons and the protective effect of nimodipine.Methods The cultured newborn rat hippocampal neurons were treated with aggregated Aβ1 40. MTT staining and LSCM was used to detected the survival rate and the intracellular free calcium concentration of neurons respectively.Results The survival rate was decreased and the fluorescence intensity was increased when neurons were treated with Aβ1 40 for 24 hours. There was obvious difference between exposed group and control group. Nimodipine could attenuate this effect partly.Conclusions Aβ1 40 reduce the survival rate and increase the intracellular free calcium concentration of neurons,nimodipe possesses certainly prolection.
