Title: Over expression of telomerase activity level, p53 and proliferating cell nuclear antigen(PCNA) in gastric carcinoma and their correlation with clinical pathological feature of gastric carcinoma
Abstract: OBJECTIVE:To investgate the telomerase activity ,the expression of p53 and proliferating cell nuclear antigen(PCNA) in gastric carcinoma and its relationship with gastric carcinoma differentiation, invasion and metsatasis. METHODS: Telomerase activity levels were measured in 58 gastric carcinoma and 35 adjacent normal tissue by a modified telomeric repeat amplification protocol,and p53,PCNA protein were identified in 40 gastric carcinoma and 35 adjacent normal tissue by immunochemical staining. Telomerase activity were analyzed with clinical pathological feature and correlation between telomerase activity and the expression of p53,proliferating cell nuclear antigen(PCNA). RESULTS: Telomerase activity was present 89%(49/56) in gastric carcinoma tissue and only 11.4%(4/35) normal tissue. The p53 mutations and PCNA were present in 77.5%(31/40) and 80%(32/40) of gastric carcinoma tissue respectively.But in normal tissues expression of p53 protein and PCNA the number was 8.5%(3/35) and 14.2%(5/35) respectively.The significantly difference was found between gastric carcinoma tissue and normal tissue (P=0.0256).At the same time,no significantly difference was found between telomerase activity and the differentiation(P=0.1741) or staging(P=0.0852) of gastric carcinoma.And non-correlation was found between telomerase activity and the expression of p53 protein or PCNA (P=0.0859) . CONCLUSIONS: p53,PCNA telomerase activity play independent role in differentiation and invasion of gastric carcinoma respectively. But the cases with overexpress of p53, PCNA and telomerase activity had highly lymph node metastasis rate simultaneously.
