Title: R egulation Mechanism Study of Applying Huoxue Tongluo Decoction to Treat SANFH of V-O-A Reaction Shaft and its Gene Expression
Abstract: Objective:By observing the Huoxue Tongluo Decoction in the treatment of SANFH of V-O-A reaction shaft and its regulation mechanism of gene expression,providing theoretical basis for Tongluo Bushen Method to prevent and treat SANFH.Methods:96 Japanese big ear rabbits selected were divided into two groups. Normal group was 24 and model group was 72. Model group were randomized into 4groups(high,medium,low dose,model group),using He's manufacturing model for building SANFH model. There were five groups totally. The treatment groups were fed different doses Huoxue Tongluo Decoction suspension. The model group and the normal group were fed with distilled water. In 6and 8weeks after delivered drug,eight rabbits were randomly selected plasma VEGF,AKP quantitative analysis and detection from each groups. Results:1. In 6and 8weeks after delivered drug,compared with the normal group,the model group rabbits' plasma VEGF levels increased significantly(P0. 01). Compared with the model group,the group of Huoxue Tongluo decoction's plasma VEGF levels were significantly lower(P0.05); comparison between high,medium and low dose groups were significantly different(P0.05). 2. In drug delivery6 and 8weeks,compared with the normal group,the model group rabbits' plasma AKP levels increased significantly(P0. 01).Compared with the model group,the group of Huoxue Tongluo Decoction's plasma AKP levels decreased significantly(P 0. 01); comparison between high,medium and low dose groups were significantly different(P0. 05). Conclusion:Huoxue Tongluo Decoction can significantly reduce the content of SANFH rabbits plasma VEGF and AKP. The curative effect is positively correlated with dose and time.
