Title: Impact of Yiqi Huoxue Therapies on Expressions of Nuclear Factor-ΚB,Cyclind1 and P16 In Rats with Gastric Precancerous Lesions
Abstract: Objective: To investigate the impact of Yiqi Huoxue therapies on the expressions of nuclear factor-κB( NF-κB),cyclinD1 and p16 in rats with gastric precancerous lesions. Methods: Sixty-four SD rats were randomly divided into blank control group( n = 8) and model control group( n = 56). Gastric precancerous lesions were induced in rats by MNNG injury to the stomach. Ten model rats were randomly sacrificed for histopathological examination. The rest 46 model rats were divided into four groups: model group( n = 12),high dose of Yiqi Huoxue group( n = 12),low dose of Yiqi Huoxue group( n = 12) and Weifuchun group( n = 10). The corresponding drugs were intragastrically given for 12 weeks. The expressions of NF-κB,cyclinD1 and p16 were observed by Envision. Results: Compared with the blank control group,the expressions of NF-κBp65 /IκBα were significantly greater( P 0. 01) in the model group. After Yiqi Huoxue and Weifuchun treatment,the expression of NF-κBp65 was decreased in high lose of Yiqi Huoxue group and Weifuchun group( P 0. 05) and the expression of IκBα was increased( P 0. 01) as compared with that of the model control group. Compared with the blank control group,the expressions of cyclinD1 and p16 were significantly greater( P 0. 01,P 0. 05) in the model group. After Yiqi Huoxue and Weifuchun treatment,the expression of cyclinD1 was reduced significantly in high dose of Yiqi Huoxue group and Weifuchun group( P 0. 01,P 0. 05) and the expression of p16 in high dose of Yiqi Huoxue group was increased( P 0. 01) as compared with those of the model control group. Conclusion: Yiqi Huoxue therapies can significantly adjust the expressions of NF-κB to inhibit the level of cyclinDl and improve p16 in rats,which may be one of the mechanisms underlying the therapeutic effect of Yiqi Huoxue against gastric precancerous lesions.
