Title: Repairing peripheral nerve defect by silk protein loading nerve growth factor
Abstract: Objective To research silk fibroin carrier which loads nerve growth factor(NGF) ,as well as observe its repairing of peripheral nerve defect in animals.Methods Twenty-four SD rats were randomly assigned to experimental group(A) in which silk protein carried nerve growth factor and control group(B,C,D) ,and after 12 weeks sciatic nerve conduction velocity and action potentials amplitude were measured,regenerated nerve fibers through the conventional hematoxylin-eosin staining,also axonal regeneration and ultrastructure of myelin were observed with transmission electron microscope.Statistical methods were used to analys the correlation experiment data.Results On the twelfth week,the nerve conduction velocity and action potential amplitude of group A were significantly different(P 0.05) from group C and group D,not different compared with group B(P 0.05) .HE staining showed that the amount of regenerated nerve fibers were more and diameter was thicker in group A and B,relatively less number of nerve fibers and smaller diameter were found in group D,the least number of nerve fibers and a large number of non-myelinated nerve fiber were found in group C.Regenerated nerve fibers in group A and B could be found thicker myelin and had completed axon structure by transmission electron microscope.Regenerative nerves was thin in sheath during group D.The regenerative nerves of group C were the smallest,and axon of which showed non-integrity.Conclusions Silk fibroin carrier bearing nerve growth factor shows biocompatibility and plays positive roles effect in peripheral nerve repairing.
