Title: Experimental Research of Astragalus Polysaccharide for TGF-β_1 Expression on Secondary Cholestatic Hepatic Fibrosis Rats
Abstract: Objective: To investigate the influence of astragalus polysaccharide for TGF-β1 expression on secondary cholestatic hepatic fibrosis rats.Methods:Cholestatic hepatic fibrosis in SD male rats(n=40)were induced by ligation of bile duct,and sham control group rats(n=5)were only made choledochus segregated.1 week after BDL,rats were divided into model control group(M)and drugs intervention group randomly(Y).The oral administration for astragalus polysaccharide was lasted for 4 weeks,then rats of M group was NS.All of rats were sacrificed after operated 5 week for determination liver histology,hepatic function and hepatic hydroxyproline content(Hyp).The expression of hepatic tissue's TGF-β1 was measured by the immunohistochemistry and westernblot.Result:Compared with that of sham control group,mortality of model control group rats was 35.0% and ascites frequency was 92.3%,while hepatic function was abnormal significantly,Hyp content of liver tissue and protein expression of TGF-β1 were increased obviously in model control group.Compared with that of model control group,mortality and ascites frequency were decreased significantly and weight of spleen was lessened obviously,while hepatic function was improved significantly.Hyp content of liver tissue and protein expression of TGF-β1 were all decreased(P0.01).Conclusions:Astragalus polysaccharide can restrain the expression of TGF-β1 on liver tissue,which may be a key step in the process of its intervention and treatment of Cholestasis fibrosis.
