Title: Effects of TLR4/NF-ΚB on proliferation and apoptosis in asthmatic rat airway smooth muscle cells
Abstract: 【Objective】 To explore the effects of TLR4/NF-ΚB on proliferation and apoptosis in asthmatic rat airway smooth muscle cells.【Methods】 The asthmatic rat airway smooth muscle cells were cultured in vitro and divided into 4 groups: Negative control,TNF-α group,(TNF-α+PDTC) group,(TNF-α+ TLR4) antibody group.Detected proliferation of ASMCs by MTT minim colorimetry,detected apoptosis of ASMCs by TUNNEL,detected content of NF-ΚB protein by ELISA,detected the expression of TLR4 protein and mRNA by Western blot and RT-PCR.【Results】 The proliferation of ASMCs in TNF-α group was significantly higher than control group(P 0.05).The proliferation of ASMCs in TNF-α plus PDTC group and TNF-α plus TLR4 antibody group were significantly lower than TNF-α group respectively(P 0.01).There was no significant difference in the proliferation between the TNF-α plus PDTC group and control group respectively(P 0.05).There was no significant difference in the apoptosis rate of ASMCs between the TNF-α plus PDTC group and control group respectively(P 0.05),the apoptosis rate of ASMCs in TNF-α plus PDTC group and TNF-α plus TLR4 antibody group were significantly lower than the TNF-α group and control group respectively(P 0.01).The expression of NF-κB protein in TNF-α group was significantly higher than control group and TNF-α plus PDTC group and TNF-α plus TLR4 antibody respectively(P 0.01).There was no significant difference in the expression of NF-κB protein between the TNF-α plus PDTC group and TNF-α plus TLR4 antibody respectively(P 0.05).The expression of TLR4 mRNA in TNF-α group was significantly higher than control group and TNF-α plus TLR4 antibody respectively(P 0.05).There was no significant difference in the expression of TLR4 mRNA between the TNF-α plus TLR4 antibody and control group respectively(P 0.05),The expression of TLR4 protein in TNF-α group was significantly higher than control group and TNF-α plus TLR4 antibody respectively(P 0.01).The expression of TLR4 protein in control group was significantly higher than TNF-α plus TLR4 antibody,respectively(P 0.01).【Conclusions】 TLR4 may regulate the proliferation and apoptosis in asthmatic rat airway smooth muscle cells by NF-κB.
