Title: Effect of Leermai Capsules on Pallium Neural Apoptosis Following Late Stage Cerebral Ischemia Reperfusion Injury in Rats
Abstract: OBJECTIVE:To observe the effect of Leermai capsules(LEM) on pallium neural apoptosis in rats after focal cerebral ischemia for 2 h and reperfusion for 30 days.METHODS:The models of focal cerebral ischemia reperfusion were established rats by a method of middle cerebral artery occlusion(MCAO).The adult rats were randomly divided into sham-oper-ated group,model group,Flunarizine hydrochloride-treated group,the LEM-treated group(high and low doses respectively).The expression of Fas,Bax mRNA and rats' neural apoptosis were assessed by immunohistochemistry staining,TUNEL and RT-PCR,and the image analysis was conducted as well.RESULTS:The apoptotic cells of the model group were mainly located in the ischemic margin(penumbra) of the ischemic pallium;and the expression of Fas,Bax mRNA remained increased after cerebral ischemia reperfusion for 30 days.Compared with the model group,the expression of Fas,Bax mRNA proteins and the number of the apoptosis cells in LEM-treated group were significantly lower(P0.01).LEM obviously reduced the expression of Fas and Bax mRNA in the injured side of brain,inhibited apoptosis and lessened damage of neural cells in pallium of rats after cerebral ischemia reperfusion.CONCLUSION:LEM has certain interventional effect on neural apoptosis in rats after focal cerebral ischemia reperfusion for 30 days.
