Title: In Vitro Culture and Rapid Propagation of Juglans sigillata
Abstract: Single bud stem of superior individual Juglans sigillata cv.'Nayong-1' were used as explants to establish in vitro culture and rapid propagation system.Results showed that the DKW medium supplemented with 1.0 mg·L~(-1) 6-BA+3.0 g·L~(-1) AC was suitable for germination of axillary buds.The DKW medium with 1.0mg·L~(-1) 6-BA+0.02 mg·L~(-1) IBA was optimal for the shoots multiplication,the proliferation coefficient was up to7.33 after culturing for 40 days.Both shoot tip and single axillary bud could be used for multiplication culture.One step method(low IBA concentration sustained induction) was not suitable for rooting culture.The highest rooting rate(71.73%) was obtained by two steps method,firstly the plantlets was cultured on 1/4DKW medium with 5.0 mg·L~(-1) IBA in the dark condition for 12 days,and then transferred to the 1/4DKW medium without IBA under light(3 g·L~(-1) AC and 20 g·L~(-1) sucrose).It was found that the IBA concentrations,time of dark induction,sucrose concentrations and AC contents were all related to the effect of rooting.After acclimation in pearlite and nutrient soil for 60 days,87.50%plantlets were survival.
