Title: Establishment of Regeneration System for Populus ×euramericana cv.'2001'
Abstract: Stem,leaf,and leafstalk developed from tender branches of Populus×euramericana cv.'2001' in spring were used as explants to establish tissue culture and regeneration system by supplementing hormones with different combinations in MS and WPM media to examine the optimal conditions for the induction of adventitious buds,and elongation and rooting of multiple shoots.It was found that induction of differentiation in medium MS was better than in WPM.The best results of differentiation of adventitious buds were achieved in MS medium supplemented with TDZ 0.025 mg·L-1,6-BA 0.05 mg·L-1,NAA 0.05 mg·L-1,and sucrose 30.00 g·L-1,by which the induced differentiation rates were over 90% for stem and leafstalk,60% for leaf.Best results of stem elongation were achieved after the multiple buds were imported to the MS supplemented with KT 0.25 mg·L-1,GA3 1.00 mg·L-1 and fructose 30.00 g·L-1.More than 6 shoots with a length of about 4 cm were elongated from the induced buds of leaf explants after 20-day culture.The optimal subculture medium and root inducement culture medium for shoot were 1/2 MS + IAA 0.02 mg·L-1+ IBA 0.02 mg·L-1+ AC 3.00 g·L-1 + sucrose 30.00 g·L-1,and the rooting rate was 90%.
