Title: Observation on effect of curcumin on the proliferation and apoptosis of human tongue carcinoma cell line Tca8113
Abstract: Objective To inverstigate the effect of curcumin on proliferation and apoptosis of human tongue carcinoma cell line Tca8113.Methods Human tongue carcinoma cell line Tca8113 was incubated with different concentrations of curcumin, cell proliferation was examined by MTT colormetric assay.Apoptosis was analyzed by flow cytometry(FCM), and apoptosis related protein Fas, p53, bcl-2 were examined by immuno-histochemical staining.Results Administration of 10, 30, 50, 70 μmol / L curcumin could significantly inhibit human tongue carcinoma cell line Tca8113 in a dose-dependent manner.After the cells being treated with curcumin at different concentration(30, 40, 50 μmol / L) for 24 hours, the apoptosis indexs(%) were(55.25 ± 4.78),(66.34 ± 5.43) and(84.36 ± 6.56) respectively, which were significantly higher than that of control group(6.57 ± 0.74, P 0.05) and in dose-depended manner.In the immunohistochemical examination, cells were treated with curcumin of different concentrations(30, 40, 50 μmol / L for) for 24 hours, the positive apoptosis cells rates of Fas, p53, bcl-2(%) were(56.25 ± 3.75, 64.72 ± 5.86, 78.64 ± 8.47)%,(51.78 ± 4.58, 65.15 ± 6.02, 84.32 ± 7.65)%,(21.36 ± 3.24, 18.92 ± 2.65, 12.75±2.54)% respectively,significantly higher thanthat of control group(3.26 ± 0.32)%,(2.18 ± 0.24)%,(35.54 ± 5.36)%,(P 0.05).Conclusion Curcumin can significantly inhibit human tongue carcinoma cell line Tca8113′s proliferation and can induce its apoptosis in dose-depended manner.
