Title: Significance and effect of mild hypothermia on the LN and the MMP-9 expressions after focal cerebral ischemia and reperfusion in rats
Abstract: Objective To study the expression of LN and MMP-9 during mild hypothermia.To provide experimental evidence for the relationship of LN,MMP-9 expression change and the neuroprotection of mild hypothermia.Methods We prepared models of rat focal cerebral ischemia and reperfusion.We devided them into normal control group,sham-operation group,normothermia ischemia/reperfusion group and hypothermia ischemia/reperfusion group.The two ischemic groups were further divided into 6 groups(3h,6h,12h,24h,72h,7d reperfusion,respectively).We detected the expression of LN and MMP-9 by immunohistochemistry.Results Compared with the sham-operation group,ischemia and reperfusion significantly decreased the expression of LN in capillaries after 12 hours,increased it greatly after 72 hours.Compared with normothermia group,hypothermia increased the expression of LN in penumbra significantly on same time(expect 72h,7d).Compared with the sham-operation group,ischemia and reperfusion greatly increased the expression of LN in neuron after 3 hours,decreased it greatly after 6 hours,increased it after 72 hours.Compared with normothermia group,hypothermia increased the expression of LN(except 72h) in penumbra significantly on same time.Ischemia and reperfusion greatly increased the expression of MMP-9 after 6 hours,decreased it after 12 hours,then increased it after 7 days.Compared with normothermia groups,mild hypothermia decreased the expression of MMP-9 on 3h,6h in penumbra significantly,increased it on 12h,24h,72h in penumbra significantly and then decreased it on 7d in penumbra significantly.Conclusion The LN and the MMP-9 may have relasionships with the formation and the development of the vasogenic brain edema.Mild hypothermia protects the brain by increasing the expression of LN and decreasing the expression of MMP-9 during early reperfusion period.
