Title: Preparation and release study of 5-fluorouracil loaded PLGA-Nanoparticles in vitro
Abstract: OBJECTIVE To prepare 5-fluorouracil loaded nanoparticles using the biodegradable materials-poly(L-lactic-co-glycolic acid)(PLGA),and to study the release behavior of the nanoparticles in vitro.METHODS 5-fluorouracil loaded PLGA nanoparticles were made by double emulsion(W/O/W)-liquid drying process with poly(L-lactic-co-glycolic acid)(PLGA) as carrier materials.High press homogenizer with a view was used to obtain nanoparticles with a very high grade of monodispersity.The mophology of the nanoparticles was observed,while mean diameter,size distribution,drug loading and incorporation efficiency were examined.The drug release behavior in vitro and its influencing factors were determined.RESULTS 5-Fluorouracil loaded PLGA nanoparticles were sphere like with the mean size of 85.4 nm,the drug loading of 12.4%±0.7% and the encapsulation efficiency of 64.1%±5.3%.The in vitro release properties could be expressed by the Higuchi's equation: Q=0.0585t~(1/2)+0.0874(r=0.9923).CONCLUSION 5-fluorouracil can be encapsuled in PLGA nanoparticles which showed significant sustained release.
