Title: Expression and significance of E-cadherin in rat model of hepatic fibrosis
Abstract: Objective To approach the expression of E-cadherin in rat hepatic fibrosis model,and to provide guideline for clinical diagnosis and treatment of hepatic fibrosis and cirrhosis.Methods Total 60 SD male rats were used in this experiment,among them 40 rats were administered subcutaneous injection of CCl4,2 ml/kg,twice a week,while other 20 SD rats(as the normal control group) were given injection of normal saline.Based on the duration of CCl4 injection,20 SD rats which received CCl4 injection for 7 weeks were enrolled into hepatic fibrosis group and 20 SD rats which received subcutaneous injection of CCl4 for 12 weeks were enrolled into hepatic cirrhosis group.Finally,all specimens were collected and divided into 5 groups according to hepatic fibrosis 0-Ⅳ stages.The expression of E-cadherin was determined by methods of immunohistochemistry and RT-PCR.Results A small quantity of E-cadherin was detected in specimens of 0 stage.Significantly more expression of E-cadherin was detected in specimens of Ⅲ and Ⅳ stages as compared with those in specimens of 0 stage(P0.05 or P0.01).Conclusion Expression of E-cadherin in rat hepatic fibrosis tissue increases.The quantity of expression of E-cadherin parallels the degree of hepatic fibrosis.Hence,E-cadherin has an intimate correlation with the occurrence and development of hepatic fibrosis.
