Abstract:Objective To analyze characterization of rpoB mutation in Beijing genotype of Mycobacterium tuberculosis.Methods The 102 Clinic isolates of Mycobacterium tuberculosis were analyzed by IS6110-RFLP and ...Objective To analyze characterization of rpoB mutation in Beijing genotype of Mycobacterium tuberculosis.Methods The 102 Clinic isolates of Mycobacterium tuberculosis were analyzed by IS6110-RFLP and Spoligotyping for genotyping.RpoB gene mutations of rifampin resistance in mycobacterium tuberculosis were detected by reverse-line blot hybridization.Results 73.5%(75/102)of Clinic isolates of Mycobacterium tuberculosis were Beijing genotype.55 of 62 rifampin-resistant isolates had rpoB gene mutations.Codon 526 had the highest mutational frequency.Conclusion Although no statistical different characterization of rpoB mutations were found in strains with different genotypes,but the ratio of resistance to rifampin and the ratio of rpoB mutations were statistical different between Beijing genotype and other genotype strains.Reverse-line blot hybridization assay is useful in rapid primary screening rifampin resistance strains in M.tuberculosis with high sensitivity and specificity.Read More
