Title: Molecular analysis of rpoB gene mutation in multidrug- resistant tuberculosis( MDR- TB) isolates from Nanyang
Abstract: Objective To reveal the molecular characteristics of rpoB gene mutation associated with rifampicin resistance in multidrug- resistant tuberculosis in Nanyang. Methods One hundred and seventy- two multidrug- resistant tuberculosis( MDR- TB) strains were involved to amplify the rpoB gene at rifampin resistance determining region( 81 bp) by PCR,then the PCR products were sequenced. Results Mutations in the rpoB were identified in 91. 2% of the isolates( 157 /172). Nucleotide substitutions in codons 531,526,533( 41. 3%,11. 6% and 10. 5%,respectively) were the most prevalent mutations. Conclusion Refampin resistance was mainly caused by mutation of the 81- bp region of rpoB gene in mycobacteria tuberculosis isolates from Nanyang.
