Title: Study on Tissue Culture and Tube Seedlings' Propagation of Ribes fasciculatum
Abstract: [Objective] To conduct tissue culture of Ribes fasciculatum and study propagation of tube seedlings. [Method]With the method of tissue culture,the tender stems of Ribes fasciculatum were used as materials to do the research on callus inducement,propagation and differentiation; cultivation,transplantation of tube seedlings and stable planting. [Result] The results demonstrated that MS + sucrose 35 g/L + 0. 4 mg /L NAA + 0. 4 mg/L ZT + 1. 8 mg/L 2,4-D was the ideal medium for callus inducement; MS + 1. 6 mg/L 2,4-D + 0. 6 mg/L 6-BA + 0. 3 mg/L NAA was the ideal medium for callus multiplication; MS + 0. 5 mg/L ZT + 0. 1 mg/L NAA was the optimum medium for callus differentiation cultivation and adventitious buds cultivation; The ideal medium for cultivation and propagation of tube seedlings was 1 /2MS + 0. 6 mg/L IAA + 0. 1 mg/L NAA + 16 g/L sucrose. The transplanting survival rate of tube seedlings was 96. 7% and stable planting survival rate was 98. 6%. [Conclusion]The flowering of the tube seedlings stable planted the second year can be normal and it maintained all botany characteristics of the wild Ribes fasciculatum.
