Title: Protective Effect of Danggui Buxue Decoction in Rats with Diabetic Foot
Abstract: Objective:To explore the protective mechanism of Danggui Buxue Decoction(DBD),a traditional Chinese drug which consists of radix astragali and angelica sinensis,in rats with diabetic foot.Methods:The rat model with diabetic foot was made by intraperitoneal injection of 55 mg/kg streptozotocin(STZ)and temperature control.Rats were treated with DBD or cilostazol.After 3weeks of treatment,blood glucose,lipid,glycohemoglobin(GHb),C-peptide(CP)and C-reactive protein(CRP)were measured,and gangrene of feet was also observed.RT-PCR technique was used to observe the expression of iNOS mRNA.Results:The diabetic rats induced by STZ had abnormal blood levels of glucose,lipid,GHb,CP,and CRP(P0.01or 0.05),and different degrees of gangrene.DBD alleviate all these changes(P0.01or 0.05),while cilostazol only lowered blood CP and CRP levels(P0.01).The level of the expression of iNOS mRNA was obviously higher in diabetic rats(P0.01),but DBD and cilostazol could decrease it (both P0.01).Conclusion:DBD may alleviate the disorder of blood glucose and lipid,lowerthe levels of GHb,CP and CRP,inhibit the expression of iNOS mRNA.These effects may slow down the progress of diabetic foot.
