Abstract:Objective To analyze the relationship between rifampin(RFP)-resistant phenotype and rpoB gene mutations in clinical isolates of Mycobacterium tuberculosis(MTB),and to expound characterization of rpoB ...Objective To analyze the relationship between rifampin(RFP)-resistant phenotype and rpoB gene mutations in clinical isolates of Mycobacterium tuberculosis(MTB),and to expound characterization of rpoB gene mutations in RFP-resistant isolates.Methods The RFP suscepitibility testing and rpoB gene sequences of 387 clinical MTB isolates were determined by proportion method(PM) on Lowenstein-Jensen media and PCR-sequencing,respectively.Minimal inhibition concentrations(MICs)in MTB isolates that bear mutations at codon 511 and 533 were detected.Results Of 387 clinical MTB isolates detected by PM,198 were RFP-resistant,and 189 RFP-sensitive.DNA sequencing of 198 RFP-resistant isolates showed that 192(96.97%) had mutations in rpoB gene touching upon 16 single codon mutations and 24 combination mutations of double codons.Rates of single codon mutations and combination mutations of double codons were 84.34%(167/198) and 12.63%(25/198),respectively.New mutation types of 1 single codon and 21 combination in double codons were found.The most common mutations occurred at codon 531(51.01%),526(21.21%) and 516(7.07%).Among 189 RFP-susceptible isolates,174(92.06%) had no mutation,15(7.94%) displayed mutations including 7 CTG511CCG(Leu→Pro)and 6 CTG533CCG(Leu→Pro).MIC results pointed out that mutations at codon 511 and 533 were associated with low level RFP-resistance in MTB.Conclusion The data and new discoveries obtained by this study provide theoretical and applicable basis for researching further into RFP-resistant mechanism of MTB and developing new rapid molecular drug susceptibility testing methods.Read More
