Abstract:The retinal was studied in control(C group),2 weeks(DM1 group) and 20 weeks(DM2 group) mice after the establishment of the experimental diabetic mice model.The cell apoptosis was detected by TUNEL,ret...The retinal was studied in control(C group),2 weeks(DM1 group) and 20 weeks(DM2 group) mice after the establishment of the experimental diabetic mice model.The cell apoptosis was detected by TUNEL,retinal NT was quantified by a competitive ELISA and the expression of nNOS and iNOS were analyzed by western blot.To determine the expression of nitric oxide synthase(NOS) in retina of early diabetic mice and its effect on the apoptosis of retinal cells.Results: Compared to control animals,apoptosis cells were observed in ganglin cell layer in DM1 group.TUNEL positive cells in retinal increased significantly in ganglion cell layer and inner nuclear layer,also in outer nuclear layer.NT levels significantly increased in mice retinas with diabetes,and also,NT levels in diabetic mice for 20 weeks were higher than those for 2 weeks.The expression of nNOS was significantly lower in diabetic retinas than those in control mice as determined by western blot.The nNOS level of 20 weeks diabetic mice was also lower than that in 2 weeks diabetic mice.The diabetic treatment induced a significant increase of iNOS expression in mice retinas,but no significant difference was found between 2 and 20 weeks-mice.Our results suggested that NT increases in the retinas of early diabetes mice contributed to the cell apoptosis in retina,and a loss of nNOS activity might contribute to retinal cell apoptosis in experimental diabetes.Read More
