Title: Analysis of genetic diversity and relationship among some Chinese domestic ducks and wild ducks using microsatellite marker
Abstract: In this research,10 microsatellite DNA markers in duck previously isolated through dual-suppression-PCR technique were used to analyze the genetic diversity of 13 duck breeds.The genetic parameters of mean heterozygosity(H),polymorphism information content(PIC),and Nei′s genetic distance(DA) were calculated for each breed.The UPGMA clustering dendrogram was drawn as well.The results showed that 7 microsatellite polymorphic loci were identified,and were regarded as effective markers to analyze genetic diversity and phylogenic relationship among duck breeds.Fifty one alleles were detected in all the loci,and alleles in each locus ranged from 5 to 12.The mean PIC value was between 0.402 3 and 0.601 5,and the mean heterozygosity was between 0.407 0 and 0.707 0,indicating that the genetic diversity of the 13 breeds was very abundant.UPGMA dendrogram constructed based on DA genetic distance revealed that the 13 duck breeds were clustered into 3 groups.In the clustering tree,8 indigenous duck breeds in China and Cherry Valley ducks were the first group,wild ducks were the second group,and Muscovy ducks had its own branch.The result of UPGMA was consistent with the genesis,differentiation and location of the duck populations.Both Anas platyryhynchos and Anas peocilorhycha contributed to domestic duck evolution.
