Title: Effect of MMP-9 in UK thrombolysis-associated cerebral ischemia-reperfusion injury and hemorrhagic transformation after focal cerebral ischemia in rats
Abstract: OBJECTIVE To investigate the action of matrix metalloproteinase-9 in urokinase(UK)thrombolysis-associated cerebral ischemia-reperfusion injury and hemorrhagic transformation after focal cerebral ischemia in rats.METHODS Two groups of rats were studied:(1)rats that received UK after ischemia;(2)rats were subjected to focal ischemia using a mechanical method of middle cerebral arterial occlusion and offered mechanical reperfusion 2 hours after ischemia,then they received saline.The expression of matrix metalloproteinase-9 of the two groups was analyzed by immunohistochemistry.The difference of the matrix metalloproteinase-9 expression was investigated between the two groups.RESULTS(1)MMP-9 expression appeared 24 h after ischemia in two groups,UK-treated rats showed higher up-regulation of MMP-9 than that in the mechanical models of ischemia/reperfusion.CONCLUSION There are significant differences in the MMP-9 up-regulation in mechanical versus thrombolytic reperfusion after focal ischemia.UK-treated rats showed significantly higher levels of MMP-9 than the mechanical models of ischemia/reperfusion.
