Title: Optimization of Cellulase Production Conditions for Bacillus Licheniformis ws-6
Abstract: Objective: To develop further application in the feed by optimizing the production conditions of cellulase produced by Bacillus Licheniformis ws-6.Method: Cellulase activity measured by reducing sugar content,combinded with single factor test and orthogonal optimization to optimize the production conditions of cellulase.Result: The maximum activity was recorded after 48h when cultured in 1.5%glucose,0.5% carboxymethyl cellulose sodium,1.5% yeast extract,0.05% KH2PO4 and 0.5%NaCl,with 2%inoculation amount,medium volume 30mL/flask(250mL),the initial pH was 6.5,the temperature was 37℃.Meanwhile the studies showed that Na+,K+,Mn2+,Mg2+,Zn2+,Co2+ could active the enzyme,but Cu2+ inhibited it.Conclusion: The cellulase activity was improved 1.4 times compared to original bacteria,meanwhile it has certain practical value to improve fiber degradation during the feed making process.
