Title: Effect of gambogic acid on apoptosis of gastric adenocarcinoma cell line AGS
Abstract: Objective To investigate the effect of gambogic acid(GA) on proliferation and apoptosis of human gastric adenocarcinoma cell line AGS.Methods After AGS cells were treated with different concentrations of GA 0,0.4,0.8,1.2,1.6,2.0,2.4 μmol/L,the inhibition rate of cell proliferation was measured by MTT assay,the apoptosis of AGS cells was analyzed by flow cytometry,and the protein expressions of B cell lymphoma/leukemia-2(Bcl-2) and Bcl-associated X protein(Bax) were detected by Western blot.Results GA could inhibit cell proliferation,promote cell apoptosis,down-regulate protein expression of Bcl-2 and up-regulate protein expression of Bax in a dose-dependent manner.Moreover,the IC50 value was 4.05 μg/ml.Conclusion GA can significantly inhibit proliferation and enhance apoptosis of AGS cells,which may be related to the expressions of Bcl-2 and Bax.
