Title: High Frequency Shoot Regeneration System from Leaf Explants of Melon
Abstract: In order to establish an efficient regeneration system via organogenesis in thin-skin melon(Cucumis melo L.) and offer a kind of reference materials for genetic transformation of melon,the leaf explants were used and some factors included concentration of plant growth regulators,orientation of the leaf and period of dark culture were studied.The results showed that the best medium for shoot regeneration was MS added 1.0 mg/L 6-BA.When the explants were cultured in normal photoperiod,no dark treatment,with abaxial side touching the optimal medium,the buds regeneration rate was 100% with 11 shoots per leaf disc.The concentration of plant growth regulators was 0.05 mg/L6-BA during the stage of bud stretching.On the medium 1/2 MS+ IAA 0.2 mg/L,the elongated shoots rooted at the rate of 100%.
