Title: Expression and its clinical significance of miR-345 in human gastric cancer
Abstract: Objective To investigate the expression of miR-345 in human gastric cancer and its clinical significance.Methods By quantitative real time reverse transcription-PCR,expression of miR-345 in normal gastric mucosa epithelial cells and gastric cancer cell lines,paired gastric cancer tissues and adjacent tissues were detected.Correlation analysis was applied to illustrate the relationship between the level of miR-345 and the clinical-pathological features or survival prognosis in 48 patients.Potential targets of miR-345 were searched by the Pictar and Targetscan prediction program.Results Real time PCR analyses revealed that miR-345 expressions were markedly upregulated in gastric cancer cell lines and tissues.Further statistical analyses showed that miR-345 level was strongly associated with differentiation (P=0.005),TNM stage(P=0.003),depth of invasion (P0.001) and lymph node status(P0.001);the survival time of patients with high miR-345 expression being poorer;CDKN1A,BBC3 and RUNX3 may become potential target genes of miR-345.Conclusions The level of miR-345 might play an important role in human gastric cancer,and thereby might be a novel and useful prognostic marker and a potential target for human gastric cancer treatment.
