Title: The Experimental Research of Osthole Injection for Protective Effects of Donor Lungs in Combined Heart-Lung Transplantation
Abstract: Objective: To study the protective effects and the possible mechanism of Osthole injection for donor lungs in combined heart-lung transplantation.Methods: Twenty adult healthy dogs were randomly divided into two groups.In the control group,the donor lungs were flushed with low-potassium dextran(LPD)solution and preserved at 4℃ while in the experimental group,the donor lungs were flushed with low-potassium dextran(LPD)solution added with Osthole injection(40 mg/kg) and preserved at 4℃,determing the analysis of blood gas PaO2 and PaCO2 of aorta declamping 5 min and 30 min of the anaesthetized receptor and the transplated heart and lung,measuring the contents of superoxide dismutase(SOD),malon-dialdehyde(MDA) and the wet/dry ratio(W/D) in the donor lung tissues and observing the ultrastructural changes of transplanted lung tissues respectively.Results: In the experimental group,arterial blood PaO2 of animal′s was significantly higher than that in the control group at any appointed time points(P0.05,P0.01).However,arterial blood PaCO2 of aorta declamping 5 min was significantly lower than that in the control group at any appointed time points (P0.05).The concentration of SOD in the lung tissues was statistically higher in the experimental group than that in the control group(P0.01).The MDA concentration was lower in the experimental group than that in the control group and there were significant differences between them(P0.05).The W/D in the experimental group was lower than that in the control group and there were significant differences between them(P0.05).The electron microscopic examination showed the lesions in the experimental group were milder than that in the control group.Conclusion: Osthole injection can improve graft lung function,attenuate the donor lung injury significantly and increase the successful rate of combined heart-lung transplantation.
