Title: K-ras mutation and its clinical significance in patients with colorectal cancer
Abstract: Objective To investigate the mutation of K-ras gene in colorectal cancer and its correlation with clinical and pathological features. Methods A total of 148 colorectal cancer tissue samples were detected the mutation of codon 12 and codon 13 in K-ras gene with reverse transcription-PCR assay and compared with their clinicopathological features. Results The mutation rate of K-ras gene in 148 cases of sporadic colorectal cancer was 31.8%(47/148).The mutation of codon 12 and 13 of K-ras gene was not significantly different in gender, age, diameter of tumor, gross morphology, tumor location, distant metastasis or lymph node metastasis(P0.05),but was significantly different in age and clinical stage(P0.05).Conclusion Reverse transcription-PCR can accurately detect K-ras gene mutation, which is closely correlated with patient's age and clinical stage.
