Title: Effect of Glomerular Cell Apoptosis on Renal Stracture and Function in Rat with glomerulosclerosis
Abstract: Objective To study the effect of glomerular cell apoptosis on renal stracture and renal function in rat with glomerulosclerosis.Methods Chronic glomerulosclerosis was induced by intravenous injection of amycin in the rats undergoing uninephectomy,Blood urea nitrogen(BUN),serum creati- nine(Scr)and 24 hour urine protein were determined.The glomerular nuclear cell counts were performed on HE staining slices.The Apoptosis of the glomerular cells was assessed by in situ dUTP blot nick-end labeling method(TUNEL).Results The number of apoptotic glomerular cells gradually in- creased after the intravenous injeceion of amycin and peaked 12 weeks after the injection.The apoptotic cells were 1.92±0.52,3.50±0.68,10.03±1. 67 respectively the 4th,8th and 12th weeks after the injection in the expermental group.There was significantly negative correlations between the glomeru- lar cell apoptosis index and the number of nuclear cells(r=-0.7456,P<0.01).There was significantly positive correlations between apoptosis index and BUN or Scr(r=0.8543 and 0.9006 respectively,P<0.01).Conclusions The excessive apoptosis of glomerular cells occurs in the rats with glomerulosclerosis and deteriorates the glomerulosclerosis.
