Title: Influence of hepatic artery reconstruction on rat liver graft in early stage in partial liver transplantation
Abstract: Objective The aims of this study are to explore the effects of grafts injury in early stage on liver transplantation with arterized partial liver transplantation model. Methods Male Sprague-Dawley rats were chosen for donors and recipients, and divided randomly into 3 groups: whole graft group (group A),partial graft group(group B),partial graft with arterial reconstruction (group C),separate to check on the six times on 30 minites, 2, 3, 6, 12, 24 h after transplantation, respectively. Main check index. liver function in serum; Expression of TNF-α in serum sample using ELISA; Cellular apoptosis in hepatic tissue using TUNEL; Expression of TNF-α and TNF-α receptor in hepatic tissue using RT-PCR. Results Liver function:The sequence of the TB, ALT, AST peak values is group Bgroup Cgroup A (all P0.05).The expresses of TNF-α in serum ascend obviously post-reperfusion in group A、B and C, to the peak value at 3 h, and fall-off to follow, in 12 h to 24 h, they still to keep far higher than control group. The peak value order is group Bgroup Cgroup A, the peak value of group B is much higher than those of group A and C, while there isn' t significant deviation between group A and C. The apoptosis number of group B (partial graft group) at every time marking exceeds group A (whole graft group) and group C (partial graft arterialized group). The highest of group B is in the 2 h, and group A, C is in the 3 h, but has no absolute difference in the the level of mRNA; Group B goes down absolutely after 6 hours, but after 12 and 24 h, group B, C are negative in expression. The gene-product of TNF-α receptor 2 is positive in group A, B, and C, the level in group B is higher than that in group A and C. Conclusion In part of non-arterial reconstruction rat model accepted liver transplantation, the small graft is severer in injury ischemia reperfusion than that in part of arterial reconstruction rat model. The intension of expression in TNF-α has a close relation with the severity of liver-injury, it may be one of the main cytokine of injury in graft.
