Title: The effect of nitroglycerin on the apoptosis of myocardial and protein and mRNA expression of Fas gene
Abstract: Objective To study the effect of nitroglycerin on the apoptosis of myocardial and protein and mRNA expression of Fas gene in the myocardial cells during continuous ischemia and ischemia reperfusion injury. Methods Wistar rats were randomly divided into 5 groups. Ligate the anterior descending branches of the left coronary artery with slipknot to block the blood flow and reperfuse the myocardium. The apoptotic cell was labeled by TUNEL, and S-P immunohistochemical staining and RT-PCR analysis examined the changes of the protein and mRNA expression of Fas gene respectively. Results In situ apoptotic cell labeling and semiquantitative analysis revealed: compared with continuous ischemia group and ischemia reperfusion injury group, the degree of the apoptosis of the myocardial cells and the protein and mRNA expression of the Fas gene decreased significantly in nitroglycerin-administrated group. Conclusions The precondition with nitroglycerin can lessen the degree of apoptosis of the myocardial cells significantly may be by up-regulating the protein and mRNA expression of Fas gene.
