Title: Expression of VEGF-D and MMP-2 in human gastric carcinoma and their relationship with lymph node metastasis
Abstract: Objective To investigate the significance of VEGF-D and MMP-2 expression in gastric carcinoma lymphatic metastasis.Method Immunohistochemical SABC(Strept Avidin Biotin Complex) method was used to examine the expressions of the VEGF-D and MMP-2 proteins.Result In normal gastric mucosa, staining for VEGF-D protein was negative, but in gastric carcinoma the positive rate of VEGF-D was 41.3%(P0.01).In normal gastric mucosa, positive staining for MMP-2 protein was 15%,while in gastric carcinoma was 54.0%(P0.01).In lymph node metastasis positive and negative group, the positive rates of VEGF-D in the two groups were 61.3% and 21.9% respectively, and there was significant difference between the two groups;the positive rates of MMP-2 in the two groups were 77.4% and 31.3% respectively, and there was significant difference between the two groups. The expressions of VEGF-D,MMP-2 were associated with the histological grade and the depth of gastric carcinoma invasion. And significant correlation was found between the expression of VEGF-D and MMP-2 in gastric carcinoma. Conclusion Over-expression of the two proteins may participate the lymph node metastasis of gastric carcinoma.Over-expression of VEGF-D can enhance the aggressive behavior of gastric carcinoma, and this may be very helpful for the gastric carcinoma cells to metastasis to the lymph nodes.
