Title: Screening of genes differentially expressed in NSCLC A549 resistance to DDP by cDNA micorarray
Abstract: OBJECTIVE:To screen the differential genes between the human NSCLC cell line A549 with and without resistance to DDP (A549/DDP and A549) by cDNA microarray, and offer some information to investigate the DDP resistance mechanism. METHODS: The total RNA was extracted from the cell line A549/DDP and A549, and then purified. The cDNA was obtained by reverse transcription polymerase chain reaction (RT-PCR), and then labeled with Cy5 and Cy3 fluorescence as probes, which were hybridized with cDNA microarray containing 17 101 human 16KcDNA. Subsequently, the signal images were scanned by Agilent fluorescence scanner and analyzed by Pix QuantArray software. RESULTS: Multiple genes take part in the resistance to DDP of A549. The 24 gene differentially expressed may have important position in the occurrence and development of the resistance to DDP of A549.
