Title: Protective effects of rhodioside derivative N03 on hypoxia/reoxygenation-induced injury in neonatal rat cardiomyocytes
Abstract: Objective:To investigate the effect of rhodioside derivative N03,1-C-β-D-gluco-pyranosyl-N-(2'-chlorophenyl) methyl methanamine,on the injury induced by hypoxia/reoxygenation in the cultured neonatal rat cardiomyocytes.Methods: Ventrical myocytes from neonatal rats(1-2 days) were cultured,and pretreated with various concentration of N03(2,10,50mg·L-1) before exposure to hypoxia/reoxygenation.Lactate dehydrogenase(LDH) in cell supernatant,creatine kinase(CK),AST(aspartate transaminase),SOD(superoxide dismutase) level,intracellular ATP(adenosine triphosphate),intracellular reactive oxygen species(ROS) level and Ca2+ were determined by colourimetry,luminometer and fluorescence probe,respectively.Results: N03 reduced LDH,CK and AST release and increased the activity of SOD after hypoxia-reoxygenation.It markedly attenuated the reduced elevation ATP and ROS,and Ca2+ overloading induced by hypoxia/reoxygenation.Conclusion: N03 can protect rat cardiomyocytes from the injury induced by hypoxia/reoxygenation,suggesting a potential for the treatment of myocardial ischemia.
