Abstract:Objective To explore the different contribution of resveratrol(Res)to the proliferation and apoptosis of glioma U251,U87 and SHG-44 cells in vitro,verify the apoptosis of U251 cell caused by Res.Metho...Objective To explore the different contribution of resveratrol(Res)to the proliferation and apoptosis of glioma U251,U87 and SHG-44 cells in vitro,verify the apoptosis of U251 cell caused by Res.Methods MTT assay was used to measure it's effects on the proliferation of U251,U87 and SHG-44 cells cultured with different concentrations of Res for 24h and it's effects on the proliferation of U251,U87 and SHG-44 cells for 6h,24h and 48h.The Annexin V-FITC and PI assay by flow cytometer were applied to detect the apoptosis of U251,U87 and SHG-44 cells.HE staining,Hoechst 33342 fluorescence staining,transmission electron microscope(TEM)was used to observe the changes of U251 cellular morphous.The cell cycle change of U251 cells was detected by FCM.Results Res obviously suppressed the proliferation of U251,U87 and SHG-44 giloma cells(P0.01),and the suppressive effect of U87 cell was more significant than that of U251 and SHG-44 cells;Res elicited a concentration-and time-dependent inhibition of U251 cells proliferation;Res induced cellular apoptosis of the U251,U87 and SHG-44 cells in a concentration-dependent manner,and the apoptosis rate of U87 cell was higher than that of U251 and SHG-44 cells.Res caused U251 cell cycle mainly with G1 arrest.Conclusion Res caused U87 cells more significant cytotoxicity and apoptosis than U251 and SHG-44 cells,Res induced U251 cell cycle mainly with G1 arrest.Read More
