Title: EFFECT OF AZIDE SODIUM IN MUTAGENESIS ON PHYSIOLOGICAL TRAITS OF Phalaenosis PROTOCORM-LIKE BODY in vitro
Abstract: The lethal and mutagenic effects of azide sodium on protocorm-like body(PLBs) of Phalaenosis was investigated.In these tests,PLBs of Phalaenosis were soaked for 6 hours in 2,4,6,8mmol /L azide sodium solution,and then inoculated to the culture medium.Proline,MDA,soluble sugar content,SOD and catalase activities of these cutured PLBs were measured.The results showed that,SOD activities,proline and MDA contents increased with the increase of azide sodium concentration,but catalase activities and soluble sugar content decreased.The half lethal dosage of azide sodium for Phalaenosis PLB mutagenesis was between 4 and 8mmol /L,and azide sodium at 4mmol /L had no significant physiological damage to PLBs of Phalaenosis.Thus,the optimal mutation dose for Phalaenosis PLB mutagenesis is 4mmol /L NaN3 and 6h soaking.
