Title: Mechanism of Inhibitory Effect of Sodium Bisulfite on Glycolate Oxidase Activity in Rice (Oryza sativa) Seedling Leaves
Abstract: The effect of NaHSO_3 on glycolate oxidaseactivity in rice seedlings was studied. It wasfound that 200~400 ppm NaHSO_3 inhibitedthe activity of glycolate oxidase by 38.6%~68.9% after 6 hrs. The inhibition increasedsignificantly after 12~24 hrs, then the activitygradually returned to the orignial level after 72hrs. Spraying with 100 ppm NaHSO_3 had noinhibitory effect on the activity of the enzyme upto 6 hrs, but induced a little increase in itsactivity after 12~24 hrs. As to the mechanism of the inhibitory eff-ect, it was found that NaHSO_3 inhibited cata-lase activity and induced the accumulation ofH_2O_2 in vivo. In vitro experiments showed thatH_2O_2 inhibited glycolate oxidase by oxidizingits essential sulfhydryl group. It was concludedthat NaHSO_3 inhibited glydolate oxidase in-directly through its inactivation effect on cata-1ase and induction of H_2O_2 accumulation inplant tissues which in turn inhibited glycolateoxidase.
