Title: Study on in situ Intestinal Absorption of Bufalin in Rats
Abstract: Objective To evaluate bufalin in situ intestinal absorption mechanism.Methods The rat single-pass intestinal perfusion technique was applied,and the gravimetry was used to correct the perfusion volume.The concentration of bufalin was determined by HPLC.The effects of absorption site,drug concentration,pH value and P-glycoprotein(P-gp) inhibitor on bufalin absorption were studied.Results The order of the drug absorption constant(Ka) and apparent absorption coefficient(Papp) was as follows:jejunumileumduodenumcolon.Drug concentration had little effect on Ka and Papp(P0.05).pH value of the drug solution affected drug absorption.There was no significance in Ka and Papp between presence of P-gp inhibitor and no P-gp inhibitor(P0.05).Conclusion Bufalin can be absorbed at all intestinal segments.The absorption of bufalin is a passive diffusion process and not affected by P-gp.
