Title: An Experimental Study on Inhibitory Effect of Baicalin on Human Hepatic Carcinoma Celline SMMC7721 in Vitro
Abstract: Objective To investigate the inhibitory effect of baicalin on the growth of human hepatic carcinoma cell line SMMC 7721 in vitro and discuss its mechanism.Methods The inhibitory fraction of SMMC 7721 cells was measured by MTT assay,and flow cytometry was used to determine the effect on cell cycle.Results ①.Baicalin in a concentration range of 4~0.005 312 5 mg/ml had remarkable inhibitory effect on the growth of SMMC 7721 in vitro,and had statistically significant difference in comparison with cell control guoup(P0.05).Tumor inhibition percentage of concentration 5~0.062 5 mg/ml had statistically significant difference in comparison with that of other concentration groups of baicalin(P0.01),and had no significant difference in comparison with DDP control group(P0.05).Concentration of 0.5 mg/ml had the highest inhibitory effect on the growth of SMMC 7721 in vitro,and the tumor inhibition rate was 63.8%.②After treatment of 0.5 mg/ml baicalin,there was a remarkable damage on SMMC 7721 cancer cell.A lot of cell fragments were observed on 60%~70% cells and a few cells shed and suspended in the medium.③Baicalin of 0.5 mg/ml had effect on arresting cell cycle of human hepatic carcinoma celline SMMC 7721 at G0/G1 phase.(Percentage of the cell at G0/G1 phase is 72.19%)Conclusion Baicalin has inhibitory effect on the growth of SMMC 7721 in vitro and it has effect on cell cycle.
