Title: Effects of propofol on the expression of fos protein and LDH of cultured deonatal rat cardiomyocytes subjected to hypoxia/reoxygenation
Abstract: Objective:To observe the effects of propofol on the expression of c-Fos protein in cultured neonatal rat cardiomyocytes and the concentration of lactate dehydrogenase(LDH) in culture medium during reoxygenation. Methods:The model of original cultured cardiac myocytes of SD rat was established and cardiomyocytes were subjected to hypoxia-reoxygenation injury.Cells with cardiomyocytes were divided into 6 groups: groupⅠ as control; group Ⅱ as hypoxia-reoxygenation (hypoxia 4 hours-reoxygenation 2 hours) control ; groups Ⅲ,Ⅳ,Ⅴ and Ⅵ were treated with intelipid and different doses propofol (25 μmol·L -1 ,50 μmol·L -1 and 100μmol·L -1 ) respectively since 10 minutes before hypoxia.The concentration of LDH in culture medium at 3 hours after reoxygenation was detected. Immunohistochemistry and mean gray were used to estimate the effect of propofol on the expression of c-Fos protein at 2 hours after reoxygenation in cardiomyocytes. Results:The concentration of LDH in group Ⅱwas higher than that in groupsⅠ, Ⅴand Ⅵ (P0.05), the mean gray values increased more in groupⅤ and groupⅥ than in group Ⅱ(P0.05).Conclution:Propofol decreased the release of LDH induced by hypoxia- reoxygenation and 50 μmol·L -1 and 100 μmol·L -1 Propofol could repress effectively the expression of c-Fos protein in cultured cardiomyocytes during reoxygenation. Propofol protected cultured cardiomyocytes subjected to hypoxia-reoxygenation injury.
