Abstract:Seven primer combinations with clear and high polymorphic bands have been screened from 64 EcoRI/MseI primer combinations(MseI fluorescent labeled).The genetic relationships and specific germplasms of...Seven primer combinations with clear and high polymorphic bands have been screened from 64 EcoRI/MseI primer combinations(MseI fluorescent labeled).The genetic relationships and specific germplasms of 29 ancient germplasms of Ginkgo biloba from Shandong province were analyzed.7 primer combinations generated 1003 bands,138 specific loci(including 36 absent bands,102 monomorphic bands),915 polymorphic bands,the percentage of polymorphic bands(PPB) was 91.21%,and the identification percentage per primer combination reached 100%.Genetic similarity coefficient(SC) among ancient germplasms of Ginkgo biloba varied from 0.43 to 0.84,and all germplasms were divided into four clusters when SC was at 0.71,the cluster wasn't competently consistent with geogphical distance of germplasms.According to the comprehensive analysis based on specific loci,similarity coefficient,clustering result and so on,10 germplasms(Rencheng',Jiaozhou',Laoshan2',Zhucheng2',Anshan1',Anshan2',Mengyin',Tancheng1',Tancheng2',Yeziyinxing1') were considered as importantly specific germplasms of ancient Ginkgo biloba in Shandong.Read More
