Title: Protective effect of D-polymannuronic sulfate on vascular endothelial cell injured by hydrogen peroxide
Abstract: Aim To study the effects of D-poly mannuronic sulfate(DPS) on the H_2 O_2(200μmol·L~(-1)) injured human umbilical vein endothelial cell lines(ECV304). Methods ECV304 viability was assessed by MTT assay. Lipid peroxidation in ECV304 which was monitored as MDA contents, the release of intracellular LDH and SOD level of cultured medium was assessed by kit. Moreover, intracellular concentration of Ca~(2+) and apoptosis of ECV304 were detected by flow cytometry. Results The results showed that DPS promoting the proliferation of ECV304 injured by H_2O_2. DPS of 0.1, 1 μg · mL~(-1) raise SOD level, reduced the contents of MDA and prevented the increase in release of LDH at four doses. By flow cytometry, results showed that DPS 1 μg · mL~(-1) decrease the concentration of Ca~(2+) and restrained the apoptosis of EVC304 induced by H_2 O_2. Conclusion DPS exerts protective action on injured ECV304 and its mechanism of action can be related to the anti-lipid peroxidation, the fall of the intracellular Ca~(2+) level and restraining apoptosis of ECV304.
