Title: Research on the Medium Optimization for the Plant Regeneration in vitro of Health Family
Abstract: [Objective] The basic technical reference for the rapid propagation and the plantlet regeneration of somatic hybridization of health family-Rhododendron was provided through the study on the best medium formula for its plant regeneration be means of the system of tissue culture.[Method] The Rhododendron young leaf as explants was inoculated in vitro on the 3 basic types of media: MS,WMP and 6,7-V with the different combinations of hormones,the different amounts of phosphorus and other processing and there were four repetitions in each treatment so that the de-differentiation and re-differentiation effect from these treatments were analyzed.[Results] It was found that the medium——WMP was conducive to the callus induction from leaf; the best basic medium for the differentiation of adventitious bud was 6,7-V; the medium for its subculture and multiplication was 6,7-V + ZT 0.5 mg/L + NAA 0.1 mg/L + IAA 0.1 mg/L,with the average multiplication coefficient of up to 12.25; the best rooting medium was 1/2 6,7-V + NAA 0.1mg/L + IAA 0.1mg/L,with the average rooting rate reaching to 91.20% for 30 days’ culturing.It was also found that amount-increasing of phosphorus in the medium was favor to the elongation of regeneration bud.[Conclusion] A prominent role in induction of the adventitious buds from Rhododendron young leaf culturing was played by the medium——6,7-V and the optimal regeneration system of Rhododendron culturing in vitro was established.
